Denise is a transgender woman who was born as Malcolm Lockwood. She decided to transition around nine years before the events of Season 2. Her family obviously initially did not accept her decision to transition, which caused her to become estranged from them for a long time. Her sister Maggie took it especially hard because "she missed her brother." Denise and her family eventually made up and she is first seen during Natural History, attending a large family barbecue. She resides in Dallas.

When Denise crashes her car leaving the barbecue due to loss of vision, Maggie and Will Halstead take her to Med. Denise initially hides her status as a transgender woman, but it is revealed when an echo reveals her prostate and she turns out to have prostate cancer. She remarks on the irony of the one thing she did not decide to remove during her transition now giving her cancer. While initially intent on returning to Dallas to seek treatment, Maggie and Will persuade her to stay in Chicago. For his help and sensitivity, Maggie awards Will the title of "Honorary Member of the Lockwood Tribe."

During Forever Hold Your Peace Denise returns to Chicago with her boyfriend Trevor. Initially he appears to be a gentleman, but Will sees him physically abusing Denise in the parking lot of the hospital. She has a broken lip but refuses to come inside to get checked out, but when she faints Will brings her in in a wheelchair, to Maggie's shock. They try to hide the situation from Maggie, but when she finds out Maggie conscripts Will to pay a visit to Trevor's workplace and they threaten him to never come near Denise again. When Denise realizes what they have done, she storms out of the hospital furious, and Maggie laments that she has probably lost her sister for good this time.
